TFIID is a general transcription factor which initiates preinitiation complex assembly through direct interaction with the TATA promoter element. It is a multisubunit complex consisting of a small TATA-binding polypeptide andother TATA-binding protein (TBP)-associated factors (TAFs). Although native TFIID can mediate both activator-independent (basal) and activator-dependent transcription in reconstituted systems, TBP can mediate only basal transcription. TAF II p100 (TBP-associated factor II100), also known as TAF5 or TAFII100, is the third largest subunit of human TFIID. It contains six WD40 repeats at the C-terminus and has an N-terminus capable of forming a flexible dimer. TAF II p100 plays an important role in forming the scaffold that is crucial for the assembly of the TFIID complex. TAF II p100 may also be involved in the stabilization of TAF interactions.

NCBI Description

Initiation of transcription by RNA polymerase II requires the activities of more than 70 polypeptides. The protein that coordinates these activities is transcription factor IID (TFIID), which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TAFs may participate in basal transcription, serve as coactivators, function in promoter recognition or modify general transcription factors (GTFs) to facilitate complex assembly and transcription initiation. This gene encodes an integral subunit of TFIID associated with all transcriptionally competent forms of that complex. This subunit interacts strongly with two TFIID subunits that show similarity to histones H3 and H4, and it may participate in forming a nucleosome-like core in the TFIID complex. TAF5: TAFs are components of the transcription factor IID (TFIID) complex, PCAF histone acetylase complex and TBP-free TAFII complex (TFTC). TAFs components-TIIFD are essential for mediating regulation of RNA polymerase transcription. TAF5/TAFII100 interacts strongly with the histone H4-related TAF6/TAFII80 and the histone H3-related TAF9/TAFII31, as well as a stable complex comprised of both TAF5/TAFII80 and TAF6/TAFII31. Apparently weaker interactions of TAF5/TAFII100 with TBP, TAF1/TAFII250, TAF11/TAFII28, and TAF12/TAFII20, but not TAF7/TAFII55, also have been observed. Belongs to the WD repeat TAF5 family. 2 isoforms of the human protein are produced by alternative splicing.
