Immunofluorescence (IF) (Fluorescent confocal image of HeLa cells stained with SYVN1 (HRD1) (C-term) antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AP2184a SYVN1 (HRD1) (C-term) primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min).)

Related Product Information for anti-SYVN1 antibody
HRD1 is a ubiquitin ligase whose expression is induced by the unfolded protein response (UPR) following endoplasmic reticulum stress. Expression of HRD1 protects cells from apoptosis by inducing degradation of abnormally processed proteins that accumulate in the endoplasmic reticulum. HRD1 is expressed in many tissues, strongly expressed in brain, pancreas, liver, kidney and skeletal muscle. Amano T, et al. reported that Synoviolin/Hrd1 (expressed in rheumatoid synovium) is a novel causative factor for arthropathy by triggering synovial cell outgrowth through its antiapoptotic effects. HRD1 contains one ring-type zinc finger.

NCBI Description

This gene encodes a protein involved in endoplasmic reticulum (ER)-associated degradation. The encoded protein removes unfolded proteins, accumulated during ER stress, by retrograde transport to the cytosol from the ER. This protein also uses the ubiquitin-proteasome system for additional degradation of unfolded proteins. Sequence analysis identified two transcript variants that encode different isoforms. Product Notes

The SYVN1 (Catalog #AAA6353116) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SYVN1, CT (SYVN1, HRD1, KIAA1810, E3 ubiquitin-protein ligase synoviolin, Synovial apoptosis inhibitor 1) (AP) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's SYVN1 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC), ELISA (EIA), Western Blot (WB). IHC: 1:50-100 ELISA: 1:1,000 WB: 1:100-500 Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the SYVN1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SYVN1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
