Stat3 is a key signaling molecule for many cytokines and growth-factor receptors (1). It is required for murine fetal development (2). Stat3 is constitutively activated in a number of human tumors (3,4). It possesses oncogenic potential (5) and anti-apoptotic activities (3). Stat3 is activated by tyrosine phosphorylation at Tyr705, which induces dimerization, nuclear translocation and DNA binding (6,7). Transcriptional activation seems to be regulated by serine phosphorylation at Ser727, apparently via the MAPK or mTOR pathways (8,9). Stat3 isoform expression appears to reflect biological function: the relative expression levels of Stat3alpha (86kD) and Stat3beta (79kD) depend on cell type, ligand exposure or maturation stage of the cells (10). It is notable that Stat3beta lacks the serine phosphorylation site within the C-terminal transcriptional activation domain (8).

NCBI Description

The protein encoded by this gene is a member of the STAT protein family. In response to cytokines and growth factors, STAT family members are phosphorylated by the receptor associated kinases, and then form homo- or heterodimers that translocate to the cell nucleus where they act as transcription activators. This protein is activated through phosphorylation in response to various cytokines and growth factors including IFNs, EGF, IL5, IL6, HGF, LIF and BMP2. This protein mediates the expression of a variety of genes in response to cell stimuli, and thus plays a key role in many cellular processes such as cell growth and apoptosis. The small GTPase Rac1 has been shown to bind and regulate the activity of this protein. PIAS3 protein is a specific inhibitor of this protein.
