Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Immunohistochemistry (IHC)

(MBS626358 staining SQSTM1 in Human prostate tissue sections by Immunohistochemistry (paraformaldehyde- fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with MBS626358 (1:25) for 1 hours at 37°C. A undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.)

Cells were then incubated with MBS626358 (1:200, 2h at RT). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10ug/ml, 5 min). SQSTM1 (p62) immunoreactivity is localized to autophagic vacuoles in the cytoplasm of U251 cells.)

SQSTM1/p62 is an adapter protein which binds ubiquitin and may regulate the activation of NFKB1 by TNF-alpha, nerve growth factor (NGF) and interleukin-1. This protein may play a role in titin/TTN downstream signaling in muscle cells, and may also regulate signaling cascades through ubiquitination. This protein is involved in cell differentiation, apoptosis, immune response and regulation of K(+) channels. SQSTM1/p62 also appears to play a role in macroautophagic removal of intracellular protein aggregates. Cellular depletion studies of SQSTM1/p62 have indicated a role for association with LC3 and aggregate proteins in order to facilitate normal formation of the autophagosome.

NCBI Description

This gene encodes a multifunctional protein that binds ubiquitin and regulates activation of the nuclear factor kappa-B (NF-kB) signaling pathway. The protein functions as a scaffolding/adaptor protein in concert with TNF receptor-associated factor 6 to mediate activation of NF-kB in response to upstream signals. Alternatively spliced transcript variants encoding either the same or different isoforms have been identified for this gene. Mutations in this gene result in sporadic and familial Paget disease of bone. [provided by RefSeq]

SQSTM1: an adapter protein which binds ubiquitin, shuttling proteins to the proteasome. Co-localizes along with the E3 ubiquitin ligase, TRAF6, to aggregates from Alzheimer's disease brains but not in control brain. May regulate the activation of NFKB1 by TNF-alpha, nerve growth factor (NGF) and interleukin-1. May play a role in titin downstream signaling in muscle cells. May regulate signaling cascades through ubiquitination. May be involved in cell differentiation, apoptosis, immune response and regulation of K(+) channels. Forms ternary complexes with PKCZ and Kv-beta2 or PKCZ and GABBR3. Also interacts with KCNAB1, GABRR1, GABRR2 and GABRR3. Interacts with EBI3, LCK, RASA1, PKCZ , PKCI, NR2F2, NTRK1, NTRK2, NTRK3, NBR1, MEK5, TRIM55 and MAPKAPK5. Interacts with the proteasome subunits PSMD4 and PSMC2. Interacts with K63- polyubiquitinated MAPT/TAU. Interacts with IKBKB through PRKCZ and PRKCI. Interacts with NGFR through TRAF6 and bridges that complex to NTRK1.
