Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-human SMAD9 polyclonal antibody (1:5000) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.)
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a member of the SMAD family, which transduces signals from TGF-beta family members. The encoded protein is activated by bone morphogenetic proteins and interacts with SMAD4. Two transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Jan 2010]
Uniprot Description
Smad8: transcription factor phosphorylated and activated by bone morphogenetic protein receptor kinases. Phosphorylated Smads dimerize with collaborating Smad4 and are translocated into the nucleus, where they regulate transcription of target genes. Participates in a wide range of critical processes including morphogenesis, cell-fate determination, proliferation, differentiation and apoptosis. Two alternatively spliced products have been described.

Product Notes
The SMAD9 smad9 (Catalog #AAA8101049) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-SMAD9 Antibody, Rabbit Polyclonal reacts with Human Predicted to React with: Mouse, Rat (Species predicted to react based on 100% sequence homology) and may cross-react with other species as described in the data sheet. AAA Biotech's SMAD9 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC), Immunofluorescence (IF). WB: 1:500-1:1000 ICC/IF: 1:1500-1:50,000. Researchers should empirically determine the suitability of the SMAD9 smad9 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SMAD9, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
