May be stored at 4 degree C for short-term only. For long-term storage, aliquot and store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

KCNN3 forms one of several calcium-activated potassium channels which are activated in a voltage-independent manner and have a relatively small unit conductance and high sensitivity to intracellular calcium. Activation is followed by membrane hyperpolarization. It is thought to regulate neuronal excitability by contributing to the slow component of synaptic afterhyperpolarization. The channel is blocked by apamin. KCCN3 contains an intracellular N and C terminus and 6 highly conserved transmembrane segments. It exists as a heterooligomer composed of 4 channel subunits each of which binds to a calmodulin subunit which regulates the channel activity through calcium-binding. These channels are thought to be potential therapeutic targets for disorders such as sleep apnea or sudden infant death syndrome and for regulating uterine contractions during labor. It was also suggested that KCNN3 may contribute to age-dependent decline in learning and memory and synaptic plasticity. Also it is thought that KCNN3 channel expression in endothelial cells may be a fundamental determinant of vascular tone and blood pressure.

NCBI Description

Action potentials in vertebrate neurons are followed by an afterhyperpolarization (AHP) that may persist for several seconds and may have profound consequences for the firing pattern of the neuron. Each component of the AHP is kinetically distinct and is mediated by different calcium-activated potassium channels. This gene belongs to the KCNN family of potassium channels. It encodes an integral membrane protein that forms a voltage-independent calcium-activated channel, which is thought to regulate neuronal excitability by contributing to the slow component of synaptic AHP. This gene contains two CAG repeat regions in the coding sequence. It was thought that expansion of one or both of these repeats could lead to an increased susceptibility to schizophrenia or bipolar disorder, but studies indicate that this is probably not the case. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Feb 2011]
