Secretory Leukocyte Protease Inhibitor (SLPI) is a 14kD non-glycosylated protein produced by secretory cells in the respiratory, genital and lacrimal glands, and by macrophages, B-cells and neutrophils. The functions of SLPI are diverse. The SLPI protein inhibits protease activity at sites of inflammation, protecting tissues against protease mediated damage and it possesses anti-microbial/anti-viral activity. In addition, SLPI has been reported to exhibit anti-inflammatory properties, regulating lipopolysaccharide induced activation of NF-kappa beta in peripheral blood monocytes.

This gene encodes a secreted inhibitor which protects epithelial tissues from serine proteases. It is found in various secretions including seminal plasma, cervical mucus, and bronchial secretions, and has affinity for trypsin, leukocyte elastase, and cathepsin G. Its inhibitory effect contributes to the immune response by protecting epithelial surfaces from attack by endogenous proteolytic enzymes; the protein is also thought to have broad-spectrum anti-biotic activity.
