Specificity
Undiluted antibody shows a strong Scl-70 precipitin band. Specificity is verified by a precipitin line of identity with the CDC-AF reference serum. This product shows no reactivity at a dilution of 1:100 when tested against purified Sm/RNP, SS-A, SS-B, Sm or Jo-1 antigens in an ELISA system designed to detect all IgG subclasses. Exhibits strong positive in an Anti-Scl-70 ELISA system.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Topoisomerase I changes the topology of DNA via cleavage of single strand only. Suitable for use in DNA conformational analysis and topology as well as DNA repair, drug resistance, cell proliferation and leukemia studies. The double-helical configuration that DNA strands naturally reside in makes them difficult to separate, and yet they must be separated if enzymes are to transcribe the sequences that encode proteins, or if chromosomes are to be replicated. In so-called circular DNA, in which double helical segment is bent around and joined in a circle, the two strands are topologically linked, or knotted. They cannot be separated by any process that does not involve the breaking of strands. Topoisomerases catalyze and guide the unknotting of DNA.
