Western Blot (WB)

(Anti-RNF34 Antibody - Western Blot. Western blot of Protein A Purified anti-RNF34 antibody shows detection of human RNF34 (arrowhead) in lysate. Lanes corresponding to empty vector 293T cell lysate (mock, left); RNF34 transfected lysate (middle) and RFFL transfected lysate (right), are shown using 20 ul of lysate per lane. Lysates were prepared from equivalent numbers of cells. Data presented demonstrate that this reagent is specific for RNF34. After SDS-PAGE and transfer, the membrane was probed with the primary antibody diluted to 1:1000 using 5% BLOTTO, 0.1% Tween-20 in PBS as the diluent. Incubation occurred for 1 h at room temperature. NCBI Description

The protein encoded by this gene contains a RINF finger, a motif known to be involved in protein-protein and protein-DNA interactions. This protein interacts with DNAJA3/hTid-1, which is a DnaJ protein reported to function as a modulator of apoptosis. Overexpression of this gene in Hela cells was shown to confer the resistance to TNF-alpha induced apoptosis, suggesting an anti-apoptotic function of this protein. This protein can be cleaved by caspase-3 during the induction of apoptosis. This protein also targets p53 and phospho-p53 for degradation. Alternatively splicing results in multiple transcript variants encoding distinct isoforms. [provided by RefSeq, Feb 2012]

Uniprot Description

RNF34: Has E3 ubiquitin-protein ligase activity. Regulates the levels of CASP8 and CASP10 by targeting them for proteasomal degradation. Protects cells against apoptosis induced by TNF. Binds phosphatidylinositol 5-phosphate and phosphatidylinositol 3- phosphate. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The RNF34 rnf34 (Catalog #AAA240834) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Rabbit Polyclonal to Human RNF34 reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's RNF34 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC - Paraffin), Western Blot (WB), ELISA (EIA). ELISA (1:4000 - 1:20000), IHC-P (10 ug/ml), WB (1:500 - 1:3000) Usage: Immunohistochemistry: was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antib.. Researchers should empirically determine the suitability of the RNF34 rnf34 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RNF34, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
