Subcellular Location: Nucleus.
Subunit Structure: Binds DNA as a monomer. Interacts with CEBPD and NONO (By similarity). Interacts with RUNX1 and SPIB. Interacts with GFI1; the interaction represses SPI1 transcriptional activity. Interacts with CEBPE (PubMed:26019275).
Similarity: Belongs to the ETS family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)31kDa.
NCBI Description
This gene encodes an ETS-domain transcription factor that activates gene expression during myeloid and B-lymphoid cell development. The nuclear protein binds to a purine-rich sequence known as the PU-box found near the promoters of target genes, and regulates their expression in coordination with other transcription factors and cofactors. The protein can also regulate alternative splicing of target genes. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
Uniprot Description
PU.1: Binds to the PU-box, a purine-rich DNA sequence (5'- GAGGAA-3') that can act as a lymphoid-specific enhancer. This protein is a transcriptional activator that may be specifically involved in the differentiation or activation of macrophages or B- cells. Also binds RNA and may modulate pre-mRNA splicing. Binds DNA as a monomer. Interacts with CEBPD and NONO. Interacts with RUNX1 and SPIB. Interacts with GFI1; the interaction represses SPI1 transcriptional activity. Highly expressed in both FV-P and FV-A-induced erythro- leukemia cell lines that have undergone rearrangements of the Spi- 1 gene due to the insertion of SFFV. Belongs to the ETS family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes
The SPI1 spi1 (Catalog #AAA9612029) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The PU.1/Spi1 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Dog(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's PU.1/Spi1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the SPI1 spi1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PU.1/Spi1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
