Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Dilution: ELISA: 0.1-1ug/ml
Western Blot: 2-4ug/ml. Recognizes only the 28kD band in MCF-7 cell lysate.
Immunohistochemistry: 1-2ug/ml in formalin-fixed, paraffin embedded tissues. For best results heat induced epitope retrieval (HIER) with citrate buffer, pH 6.0, is required prior to staining.

Pituitary tumor-transforming gene 1 (PTTG-1) is an oncogene that is expressed in normal tissues containing proliferating cells and in tumors. PTTG is downstream of the MAP kinase cascade and its phosphorylation at Ser162 by MAPK in vitro. The phosphorylation plays an essential role in PTTG's transactivation function. PTTG has a DNA-binding domain, and it activates c-Myc (an oncogene) transcription by binding to the promoter in a protein complex. Studies found that expression of human PTTG correlates with cell proliferation in a cell cycle-dependent manner and the association of hPTTG with Ku, the regulatory subunit of DNA-dependent protein kinase, is disrupted when a DNA double-strand breaks occurs.
