Related Product Information for anti-PTTG1 antibody
The product of the oncogene PTTG, pituitary tumor transforming gene, is a human homolog of the anaphase-inhibitor vertebrate protein, securin. PTTG contains a basic amino-terminal domain and an acidic carboxy-terminal domain, which acts as a transactivation domain when fused to a heterologous DNA binding domain. Human PTTG is overexpressed in Jurkat and is also detected in human thymus, testis and placenta. PTTG is mainly expressed in the cytoplasm and is also partially localized to the nucleus. Vertebrate PTTG regulates the separin Esp1, which promotes chromatid separation, to overcome the cohesive forces that hold sister chromatids together. This regulatory function of PTTG suggests that defective regulation of cohesion may contribute to cancer by promoting chromosome instability. Although vertebrate PTTG shares cell-cycle functions with its yeast securin counterparts Pds1p and Cut2, none share sequence homology.

NCBI Description

The encoded protein is a homolog of yeast securin proteins, which prevent separins from promoting sister chromatid separation. It is an anaphase-promoting complex (APC) substrate that associates with a separin until activation of the APC. The gene product has transforming activity in vitro and tumorigenic activity in vivo, and the gene is highly expressed in various tumors. The gene product contains 2 PXXP motifs, which are required for its transforming and tumorigenic activities, as well as for its stimulation of basic fibroblast growth factor expression. It also contains a destruction box (D box) that is required for its degradation by the APC. The acidic C-terminal region of the encoded protein can act as a transactivation domain. The gene product is mainly a cytosolic protein, although it partially localizes in the nucleus. Three transcript variants encoding the same protein have been found for this gene. Uniprot Description

Securin: Regulatory protein, which plays a central role in chromosome stability, in the p53/TP53 pathway, and DNA repair. Probably acts by blocking the action of key proteins. During the mitosis, it blocks Separase/ESPL1 function, preventing the proteolysis of the cohesin complex and the subsequent segregation of the chromosomes. At the onset of anaphase, it is ubiquitinated, conducting to its destruction and to the liberation of ESPL1. Its function is however not limited to a blocking activity, since it is required to activate ESPL1. Negatively regulates the transcriptional activity and related apoptosis activity of TP53. The negative regulation of TP53 may explain the strong transforming capability of the protein when it is overexpressed. May also play a role in DNA repair via its interaction with Ku, possibly by connecting DNA damage-response pathways with sister chromatid separation. Belongs to the securin family.

Product Notes

The PTTG1 pttg1 (Catalog #AAA3013418) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Securin Polyclonal Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's Securin can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunocytochemistry (ICC), Immunofluorescence (IF), Immunohistochemistry (IHC), Flow Cytometry (FC/FACS), Immunoprecipitation (IP). WB: 1:500 ICC: 1:50-1:200 IHC: 1:50-1:200 FC/FACS: 1:50-1:100. Researchers should empirically determine the suitability of the PTTG1 pttg1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Securin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
