Dilution: Western Blot: 0.5-2ug/ml. Detects PP2A catalytic subunit (~34kD) in rabbit brain membrane cytosol and in human A431 RIPA cell lysate.
Immunoprecipitation: 4ug immunoprecipitates PP2A catalytic subunit from 500ug rabbit brain extract and 1mg human A431 RIPA cell lysate.
Immunoprecipitation/Phosphatase Assay: 5ug immunoprecipitates PP2A from 1U of purified Protein Phosphatase 2A, Serine, Threonine (PP2A Ser/Thr) (Catalog No: P9107-33D). Phospatase activity was demonstrated using Phosphatase Assay Kit Ser/Thr, KR-pT-IRR, BioAssay (Catalog No: P4071-70).
Optimal dilutions to be detemined by the researcher.

Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Type 2 protein phosphatases are serine/threonine phosphatases which can preferentially dephosphorylate the alpha subunit of phosphorylase kinase and are insensitive to inhibitor 2 treatment. Protein phosphatase 2A (PP2A) can be distinguished from PP2B and PP2C which require Ca2+ and Mg2+, respectively, by its partial activity in the absence of divalent cations. PP2A activity is potently inhibited by treatment with okadaic acid and shows much stronger activity toward histone phosphorylated by Protein Kinase C and casein phosphorylated by Protein Kinase A than other protein phosphatases.
