Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Protein Kinase Cd (PKCd) is a 78kD member of the novel group (nPKCs: sensitive to diacylglycerol, phosphatidylserine and phorbol esters) of the PKC family of serine/threonine kinases that are involved in a wide range of physiological processes including mitogenesis, cell survival and transcriptional regulation. PKCd is an ubiquitously expressed PKC isozyme that has been implicated in the regulation of multiple cellular processes including cell cycle progression and apoptosis. In contrast to other PKC isozymes where phosphorylation of the activation loop residue is essential for catalytic activity, phosphorylation of the activation loop threonine 507 of PKCd appears to modulate, but is not required for, catalytic activity. This phosphorylation may also regulate PKCd stability and degradation. Autophosphorylation at serine 645 (serine 643 in mouse and rat) is not required for, but may modulate catalytic activity and receptor dephosphorylation.

NCBI Description

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play distinct roles in cells. The protein encoded by this gene is one of the PKC family members. Studies both in human and mice demonstrate that this kinase is involved in B cell signaling and in the regulation of growth, apoptosis, and differentiation of a variety of cell types. Alternatively spliced transcript variants encoding the same protein have been observed. [provided by RefSeq]
