NCBI Description

The protein encoded by this gene is a zinc finger protein with histone methyltransferase activity that catalyzes histone H3 lysine 4 trimethylation (H3K4me3) during meiotic prophase. This protein contains multiple domains, including a Kruppel-associated box (KRAB) domain, an SSX repression domain (SSXRD), a PRD1-BF1 and RIZ homologous region, a subclass of SET (PR/SET) domain, and a tandem array of C2H2 zinc fingers. The zinc finger array recognizes a short sequence motif, leading to local H3K4me3, and meiotic recombination hotspot activity. The observed allelic variation alters the DNA-binding sequence specificity of the protein, resulting in distinct meiotic recombination hotspots amongst individuals and populations. Multiple alternate alleles of this gene have been described. [provided by RefSeq, Jul 2015]

Uniprot Description

PRDM9: Histone methyltransferase that specifically trimethylates 'Lys-4' of histone H3 during meiotic prophase and is essential for proper meiotic progression. Does not have the ability to mono- and dimethylate 'Lys-4' of histone H3. H3 'Lys-4' methylation represents a specific tag for epigenetic transcriptional activation. Plays a central role in the transcriptional activation of genes during early meiotic prophase.

Protein type: C2H2-type zinc finger protein; Methyltransferase; EC 2.1.1.43; Methyltransferase, protein lysine

Chromosomal Location of Human Ortholog: 5p14

Cellular Component: chromosome; nucleoplasm

Molecular Function: histone-lysine N-methyltransferase activity; metal ion binding; nucleic acid binding

Biological Process: meiotic gene conversion; regulation of transcription, DNA-dependent; transcription, DNA-dependent

Product Notes

The PRDM9 prdm9 (Catalog #AAA422052) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-PRDM9 Antibody reacts with Expected from sequence similarity: Human and may cross-react with other species as described in the data sheet. AAA Biotech's PRDM9 can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA). Peptide ELISA: Antibody detection limit dilution 1: 4000. Western Blot: Preliminary experiments gave an approx 80kDa band in Human Prostate, Testis and Skeletal Muscle lysates after 1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 103kDa according to NP_064612.2. The 80kDa band was successfully blocked by incubation with the immunizing peptide. Researchers should empirically determine the suitability of the PRDM9 prdm9 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: PDPHSRNDKT KG. It is sometimes possible for the material contained within the vial of "PRDM9, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
