On the basis of both functional and structural considerations, members of the IkappaB family of proteins can be divided into four groups. The first of these groups, IkappaB-alpha, includes the avian protein pp40 and the mammalian MAD-3, both of which inhibit binding of p50-p65 NFkappaB complex or Rel protein to their cognate binding sites but do not inhibit the binding of p50 homodimer to kappaB sites, suggesting that the IkappaB-alpha family binds to the p65 subunit of p50-p65 heterocomplex through Ankyrin repeats. The second member of the IkappaB family is represented by a protein designated IkappaB-beta . The third group of IkappaB proteins is represented by IkappaB-gamma, a protein identical in sequence with the C-terminal domain of the p110 precursor of NFkappaB p50 and expressed predominantly in lymphoid cells. An additional IkappaB family member has been identified as IkappaB-epsilon, a protein which has several phosphorylated forms and is primarily found complexed with RelA and/or c-Rel.

NCBI Description

This gene encodes a member of the NF-kappa-B inhibitor family, which contain multiple ankrin repeat domains. The encoded protein interacts with REL dimers to inhibit NF-kappa-B/REL complexes which are involved in inflammatory responses. The encoded protein moves between the cytoplasm and the nucleus via a nuclear localization signal and CRM1-mediated nuclear export. Mutations in this gene have been found in ectodermal dysplasia anhidrotic with T-cell immunodeficiency autosomal dominant disease. [provided by RefSeq, Aug 2011]
