Preparation and Storage
Lyophilized powder may be stored at -20 degree C for short-term only. Reconstitute with sterile 40-50% glycerol, aliquot and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

PDX-1 (Pancreas-duodenum homeobox 1; also IPF-1, IDX-1, STF-1, GSF and IUF-1) is a 42kD member of the IPF-1 subfamily, ANTP homeobox family of proteins. It is expressed in developing pancreas, plus pancreatic beta-cells, and duodenal crypt epithelium. In concert with HNF6, PDX-1 drives pancreatic endocrine cell formation by regulating the expression of neurogenin-3. In adult beta-cells, PDX-1 binds to PCIF-1 to regulate insulin production. Mouse PDX-1 is 284aa in length (Accession # P52946). It contains one transactivation domain (aa13-73) a PBX1 P-P-M-W-K dimerization motif (aa119-124), and a DNA-binding homeodomain with an embedded NLS (aa148-206). Full-length mouse PDX-1 shares 94% and 88% aa identity with rat and human PDX-1, respectively.

Uniprot Description

Function: Required for anchoring dihydrolipoamide dehydrogenase (E3) to the dihydrolipoamide transacetylase (E2) core of the pyruvate dehydrogenase complexes of eukaryotes. This specific binding is essential for a functional PDH complex.

Subunit structure: Eukaryotic pyruvate dehydrogenase (PDH) complexes are organized as a core consisting of the oligomeric dihydrolipoamide acetyl-transferase (E2), around which are arranged multiple copies of pyruvate dehydrogenase (E1), dihydrolipoamide dehydrogenase (E3) and protein X (E3BP) bound by non-covalent bonds.

Subcellular location: Mitochondrion matrix.

Miscellaneous: Present with 414 molecules/cell in log phase SD medium. Ref.7
