Tyrosine-protein kinase that acts ascell-surface receptor for homodimeric PDGFBand PDGFD and for heterodimers formed byPDGFA and PDGFB, and plays an essentialrole in the regulation of embryonicdevelopment, cell proliferation, survival, differentiation, chemotaxis and migration.Plays an essential role in blood vesseldevelopment by promoting proliferation, migration and recruitment of pericytes andsmooth muscle cells to endothelial cells.Plays a role in the migration of vascularsmooth muscle cells and the formation ofneointima at vascular injury sites. Requiredfor normal development of the cardiovascularsystem. Required for normal recruitment ofpericytes (mesangial cells) in the kidneyglomerulus, and for normal formation of abranched network of capillaries in kidneyglomeruli. Promotes rearrangement of theactin cytoskeleton and the formation ofmembrane ruffles. Binding of its cognateligands - homodimeric PDGFB, heterodimersformed by PDGFA and PDGFB or homodimericPDGFD -leads to the activation of severalsignaling cascades; the response depends onthe nature of the bound ligand and ismodulated by the formation of heterodimersbetween PDGFRA and PDGFRB.Phosphorylates PLCG1, PIK3R1, PTPN11,RASA1/GAP, CBL, SHC1 and NCK1. Activationof PLCG1 leads to the production of thecellular signaling molecules diacylglyceroland inositol 1,4,5-trisphosphate, mobilizationof cytosolic Ca(2+) and the activation ofprotein kinase C. Phosphorylation of PIK3R1,the regulatory subunit of phosphatidylinositol3-kinase, leads to the activation of the AKT1signaling pathway. Phosphorylation of SHC1,or of the C-terminus of PTPN11, creates abinding site for GRB2, resulting in theactivation of HRAS, RAF1 and down-streamMAP kinases, including MAPK1/ERK2 and/orMAPK3/ERK1. Promotes phosphorylation andactivation of SRC family kinases. Promotesphosphorylation of PDCD6IP/ALIX and STAM.Receptor signaling is down-regulated byprotein phosphatases that dephosphorylatethe receptor and its down-stream effectors, and by rapid internalization of the activatedreceptor

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a cell surface tyrosine kinase receptor for members of the platelet-derived growth factor family. These growth factors are mitogens for cells of mesenchymal origin. The identity of the growth factor bound to a receptor monomer determines whether the functional receptor is a homodimer or a heterodimer, composed of both platelet-derived growth factor receptor alpha and beta polypeptides. This gene is flanked on chromosome 5 by the genes for granulocyte-macrophage colony-stimulating factor and macrophage-colony stimulating factor receptor; all three genes may be implicated in the 5-q syndrome. A translocation between chromosomes 5 and 12, that fuses this gene to that of the translocation, ETV6, leukemia gene, results in chronic myeloproliferative disorder with eosinophilia. Uniprot Description
PDGFRB: a receptor tyrosine kinase of the PDGFR family that binds members of the platelet-derived growth factor family. The identity of the growth factor bound determines whether the functional receptor is a homodimer or a heterodimer, composed of both PDGFR-alpha and -beta. Ligand binding induces receptor dimerization and autophosphorylation, thereby recruiting SH2-containing proteins including Grb2, Src, GAP, PTPN11, PI3 kinase, PLC-gamma and Nck. Regulates cell growth, actin reorganization, migration and differentiation. A variety of myeloproliferative disorders and cancers result from translocations that activate PDGFRbeta by fusion with proteins such as TEL/ETV6, H2, CEV14/TRP11, rabaptin 5, and huntington interacting protein 1. Gleevec treatment of TEL fusions has been successful. Overexpressed in metastatic medulloblastoma. Inhibitors: Gleevec, Sutent.

The PDGFRB pdgfrb (Catalog #AAA9215252) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 40-72. The PDGFRB Antibody (N-term) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's PDGFRB can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistochemistry-Paraffin (IHC-P), Immunofluorescence (IF), Western Blot (WB), Flow Cytometry (FC/FACS). IF~~1:10~50 IHC-P~~1:10~50 FC~~1:10~50 WB~~1:1000. Researchers should empirically determine the suitability of the PDGFRB pdgfrb for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PDGFRB, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
