Related Product Information for anti-PARP3 antibody
The protein encoded by this gene belongs to the PARP family. These enzymes modify nuclear proteins by poly-ADP-ribosylation, which is required for DNA repair, regulation of apoptosis, and maintenance of genomic stability. This gene encodes the poly(ADP-ribosyl)transferase 3, which is preferentially localized to the daughter centriole throughout the cell cycle. Alternatively spliced transcript variants encoding different isoforms have been identified.

NCBI Description

The protein encoded by this gene belongs to the PARP family. These enzymes modify nuclear proteins by poly-ADP-ribosylation, which is required for DNA repair, regulation of apoptosis, and maintenance of genomic stability. This gene encodes the poly(ADP-ribosyl)transferase 3, which is preferentially localized to the daughter centriole throughout the cell cycle. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Involved in the base excision repair (BER) pathway, by catalyzing the poly(ADP-ribosyl)ation of a limited number of acceptor proteins involved in chromatin architecture and in DNA metabolism. This modification follows DNA damages and appears as an obligatory step in a detection/signaling pathway leading to the reparation of DNA strand breaks. May link the DNA damage surveillance network to the mitotic fidelity checkpoint. Negatively influences the G1/S cell cycle progression without interfering with centrosome duplication. Binds DNA. May be involved in the regulation of PRC2 and PRC3 complex-dependent gene silencing. Ref.7

Catalytic activity: NAD+ + (ADP-D-ribosyl)(n)-acceptor = nicotinamide + (ADP-D-ribosyl)(n+1)-acceptor.

Subunit structure: Interacts with PRKDC and PARP1. Interacts with XRCC5; the interaction is dependent on nucleic acids. Interacts with XRCC6; the interaction is dependent on nucleic acids. Interacts with EZH2, HDAC1, HDAC2, SUZ12, YY1, LRIG3 and LIG4. Ref.7

Subcellular location: Nucleus. Cytoplasm › cytoskeleton › microtubule organizing center › centrosome. Cytoplasm › cytoskeleton › microtubule organizing center › centrosome › centriole. Note: Core component of the centrosome. Preferentially localized to the daughter centriole throughout the cell cycle. According to Ref.7, it is almost exclusively localized in the nucleus and appears in numerous small foci and a small number of larger foci whereas a centrosomal location has not been detected. Ref.3 Ref.7

Tissue specificity: Widely expressed; the highest levels are in the kidney, skeletal muscle, liver, heart and spleen; also detected in pancreas, lung, placenta, brain, leukocytes, colon, small intestine, ovary, testis, prostate and thymus.

Domain: According to Ref.2, the N-terminal domain (54 amino acids) of isoform 2 is responsible for its centrosomal localization. The C-terminal region contains the catalytic domain.

Post-translational modification: Auto-poly(ADP)-ribosylation.

Sequence similarities: Contains 1 PARP alpha-helical domain.Contains 1 PARP catalytic domain.

Product Notes

The PARP3 parp3 (Catalog #AAA648982) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The PARP3, ID (Poly [ADP-ribose] Polymerase 3, PARP-3, hPARP-3, IRT1, NAD(+) ADP-ribosyltransferase 3, ADPRT-3, Poly[ADP-ribose] Synthase 3, pADPRT-3, ADPRT3, ADPRTL3) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's PARP3 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Immunohistochemistry (IHC). Suitable for use in ELISA and Immunohistochemistry. Dilution: ELISA: 1:2000 Immunohistochemistry (Formalin fixed paraffin embedded): 3.75ug/ml. Researchers should empirically determine the suitability of the PARP3 parp3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "PARP3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
