Western blot analysis (0.5-4 ug/ml). However, the optimal conditions should be determined individually. The antibody recognizes ~60-70 kDa Pak4 in samples of human, mouse, and rat origins. The antibody does not cross-react with other Paks. Reactivity to other species has not been tested.

Background: The PAK (p21-activated kinase) family of serine/threonine kinases plays an important role in multiple cellular processes, including cytoskeletal reorganization, MAPK signaling, apoptotic signaling, etc. Several mechanisms that induce PAK activation have been reported. Binding of Rac/cdc42 to the CRIB (or PBD) domain at the N-terminal region of PAK causes autophosphorylation and conformational change of PAK. More recently identified members PAK4, PAK5 and PAK6 have lower sequence similarity with PAK1-3 in the regulatory N-terminal region. It has been demonstrated that phosphorylation of serine 474 of PAK4 may play a pivotal role in the activity and function of PAK4 kinase

PAK proteins, a family of serine/threonine p21-activating kinases, include PAK1, PAK2, PAK3 and PAK4. PAK proteins are critical effectors that link Rho GTPases to cytoskeleton reorganization and nuclear signaling. They serve as targets for the small GTP binding proteins Cdc42 and Rac and have been implicated in a wide range of biological activities. PAK4 interacts specifically with the GTP-bound form of Cdc42Hs and weakly activates the JNK family of MAP kinases. PAK4 is a mediator of filopodia formation and may play a role in the reorganization of the actin cytoskeleton. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
