Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Paired immunoglobulin-like, type 2 receptor beta (PILR-b) is a type I transmembrane (TM) glycoprotein belonging to the Ig superfamily. It is the activating counterpart to the ITIM-bearing PILR-a inhibitory receptor. PILR-b is expressed in a wide variety of tissues including hematopoietic cells. Mature human PILR-b is a 208 amino acid (aa) protein with one V-type Ig-like extracellular domain, a truncated cytoplasmic tail, and positively-charged residues in its TM domain that interacts with ITAM-bearing adaptor molecules. Within the V-type Ig-like region in their ECD, human PILR-b and PILR-a share a 92% aa sequence identity. The aa sequence of mouse PILR-b ECD is only 43% identical to that of the human protein.

NCBI Description

Cell signaling pathways rely on a dynamic interaction between activating and inhibiting processes. SHP-1-mediated dephosphorylation of protein tyrosine residues is central to the regulation of several cell signaling pathways. Two types of inhibitory receptor superfamily members are immunoreceptor tyrosine-based inhibitory motif (ITIM)-bearing receptors and their non-ITIM-bearing, activating counterparts. Control of cell signaling via SHP-1 is thought to occur through a balance between PILRalpha-mediated inhibition and PILRbeta-mediated activation. These paired immunoglobulin-like receptor genes are located in a tandem head-to-tail orientation on chromosome 7. This particular gene encodes the non-ITIM-bearing member of the receptor pair, which has a truncated cytoplasmic tail relative to its ITIM-bearing partner, and functions in the activating role. Alternatively spliced transcript variants have been noted for this gene, but the full-length nature of many is not known. [provided by RefSeq]
