WB: The detection limit for Osteocalcin is approximately 0.25ng/lane under reducing conditions.

Predicted Species: Species predicted to be fit for the product based on sequence similarities.
By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections.

Other applications have not been tested.
Optimal dilutions should be determined by end users.
Immunogen
E Coli-derived human Osteocalcin recombinant protein (Position: Y52-V100). Human Osteocalcin shares 69% and 86% amino acid (aa) sequences identity with mouse and rat Osteocalcin, respectively.
Contents
Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg NaN3.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Preparation and Storage
At -20°C for one year. After reconstitution, at 4°C for one month. It can also be aliquotted and stored frozen at -20°C for a longer time. Avoid repeated freezing and thawing.

Background: Osteocalcin, also known as bone gamma-carboxyglutamic acid-containing protein (BGLAP), is a noncollagenous protein found in bone and dentin. In humans, the osteocalcin is encoded by the BGLAP gene. Its receptor is GPRC6A. It is mapped to 1q22. Osteocalcin may play a role in the body's metabolic regulation and is pro-osteoblastic, or bone-building. Osteocalcin acts as a hormone in the body, causing beta cellsin the pancreas to release more insulin, and at the same time directing fat cells to release the hormone adiponectin, which increases sensitivity to insulin. It may play a role in male fertility. It is found that picomolar amounts of osteocalcin affected insulin secretion and beta-cell proliferation.

NCBI Description

This gene encodes a highly abundant bone protein secreted by osteoblasts that regulates bone remodeling and energy metabolism. The encoded protein contains a Gla (gamma carboxyglutamate) domain, which functions in binding to calcium and hydroxyapatite, the mineral component of bone. Serum osteocalcin levels may be negatively correlated with metabolic syndrome. Read-through transcription exists between this gene and the neighboring upstream gene, PMF1 (polyamine-modulated factor 1), but the encoded protein only shows sequence identity with the upstream gene product.
