Applicable Applications for anti-NOS I antibody
Western Blot (WB)
Application Notes
Suitable for use in Western Blot.
Dilution: Western Blot: 1:500 detects affinity purified bacterial expressed rat nNOS/NOS I that had been phosphorylated by purified AMPK in vitro. Lambda phosphatase treated and untreated affinity purified bacterial expressed rat nNOS/NOS I that had been phosphorylated by purified AMPK was resolved by electrophoresis, transferred to PVDF and probed with N5350-02 (1:500). Proteins were visualized using a donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
Immunogen
Synthetic peptide corresponding to aa surrounding Ser1417 of human neuronal Nitric Oxide Synthase, with an N-terminal cysteine added for KLH conjugation purposes, where pS is phosphorylated serine corresponding to the AMPK phosphorylation site.
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Related Product Information for anti-NOS I antibody
Nitric oxide (NO), a diffusible free radical gas, acts as a neurotransmitter in brain and peripheral nervous system. It accounts for the activity of endothelium-derived relaxing factors, which stimulate vasodilatation by releasing NO from the endothelium. Unlike typical neurotransmitter, NO is a not stored in synaptic vesicle and does not act on membrane receptors. Synthesis of NO, initially demonstrated in vascular endothelium, is now found in many tissues. It is found in endothelium, adrenal glands, brain and platelets. The other principle group is inducible, Ca2+/Calmodulin-independent, and inhibited by NMMA and L-NNA. It has been found in macrophage, hepatocytes, tumor cells, vascular smooth muscle and endothelial cells. Analyses of cDNA clones have identified three distinct NOS genes in mammals: neuronal (nNOS/bNOS/NOS-I), endothelial (eNOS/NOS-III), and macrophage (mNOS/iNOS/NOS-II).

Product Notes

The NOS I (Catalog #AAA629394) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The NOS I, phosphorylated (Ser1417) (Neuronal Nitric Oxide Synthase, bNOS, nNOS, NOS-1) reacts with Human, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's NOS I can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the NOS I for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NOS I, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
