Specificity
This item specifically recognises the 180kDa NR2B subunit of the NMDA (N-methyl-D-aspartate) receptor when phosphorylated at Tyrosine 1252. Receptors for NMDA belong to a group of ionotropic glutamate receptors which play a key role in the mediation of glutamate neurotransmission within the mammalian central nervous system (CNS), including involvement in memory and learning processes. Several antagonists and agonists of NMDA receptors (NMDAR) have been identified, including the glutamate analogue Homoquinolinic acid, which displays a higher affinity for NR2B-containing NMDAR. Properties of NMDAR include modulation by glycine, inhibition by Zn2+, voltage-dependent Mg2+ blockade and high Ca2+ permeability. The involvement of NMDAR in the CNS has become a focus area for neurodegenerative diseases such as Alzheimer's disease (Popke, E.J.) and also epilepsy and ischemic neuronal cell death.

Applicable Applications for anti-NMDA antibody
Western Blot (WB)*
Application Notes
Western Blot: This item detects a band/s of approximately 180kDa in rat hippocampal cell lysates.
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Perservative Stabilisers
0.09% Sodium Azide (NaN3)
50% Glycerol
0.01% Bovine Serum Albumin
Immunogen
Phosphopeptide corresponding to smino acid residues surrounding the phosphorylated tyrosine 1252 of the NR2B subunit of rat NMDA receptor.
Buffer Solution
Antiserum Preparation
Antisera to rat NMDAR NR2B subunit pTyr1252 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG prepared by affinity chromatography.
Target Species
Rat
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of despatch.

NCBI Description

subunit of the NMDA receptor glutamate gated ion channel; displays voltage dependent sensitivity to magnesium, calcium permeability, and glycine regulated activity; may facilitate learning and long term potentiation [RGD, Feb 2006]

Uniprot Description

NMDAR2B: an NMDA receptor subtype of glutamate-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Mediated by glycine. Plays a key role in synaptic plasticity, synaptogenesis, excitotoxicity, memory acquisition and learning. Mediates neuronal functions in glutamate neurotransmission. In concert with DAPK1 at extrasynaptic sites, acts as a central mediator for stroke damage. Its phosphorylation at Ser-1303 by DAPK1 enhances synaptic NMDA receptor channel activity inducing injurious Ca2+ influx through them, resulting in an irreversible neuronal death.

Product Notes

The NMDA grin2b (Catalog #AAA221522) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI NMDA RECEPTOR NR2B (pTyr1252) reacts with Bovine (Expected from Sequence), Chicken (Expected from Sequence), Dog (Expected from Sequence), Human (Expected from Sequence), Mouse (Expected from Sequence), Zebrafish (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's NMDAR NR2B can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB)*. Researchers should empirically determine the suitability of the NMDA grin2b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NMDAR NR2B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
