Related Product Information for anti-MAP4K4 antibody
Nuclear factor kappa B (NF-kB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kB mediates the expression of a great variety of genes in response to extracellular stimuli including IL-1, TNFa, LPS and mitogens. A serine/threonine protein kinase which mediates NF-kB activation by IL-1, TNFa and CD95 was identified recently and designated NIK (for NF-kB inducing kinase) (1). NIK is an activator of IkB kinase alpha and beta (IKKa and IKKb) (2-5). Therefore, NIK is a key molecule in the NF-kB signaling pathway leading to the induction of a variety of gene expression in response to proinflammatory cytokines and bacteria products Human immunodeficiency virus (HIV) and related virus require coreceptors, in addition to CD4, to infect target cells. Some G protein-coupled receptors including CCR5, CXCR4, CCR3, CCR2b and CCR8 in the chemokine receptor family, and four new human molecules GPR15, STRL33, GPR1 and V28 were recently identified as HIV coreceptors (1,2). Among them, CCR5 (CC-CKR-5) is a principal coreceptor for macrophage- and dual-tropic HIV-1 strains fusion and entry of human white blood cells (3,4). CCR5 is required for the infection by HIV-1, HIV-2, and SIV (1,2). The b-chemokines RANTES, MIP-1a and MIP-1b are the ligands for CCR5 and prevent infection by M-tropic HIV-1 (3-5). CXC5 associates with the surface CD4-gp120 of HIV complex and leads to membrane fusion and virus entry of target cells (6,7). The amino-terminal domain and the extracellular loops of CCR5 serve as HIV biding sites (8,9). CCR5 messenger RNA is expressed in lymphoid organs and monocytes.

NCBI Description

The protein encoded by this gene is a member of the serine/threonine protein kinase family. This kinase has been shown to specifically activate MAPK8/JNK. The activation of MAPK8 by this kinase is found to be inhibited by the dominant-negative mutants of MAP3K7/TAK1, MAP2K4/MKK4, and MAP2K7/MKK7, which suggests that this kinase may function through the MAP3K7-MAP2K4-MAP2K7 kinase cascade, and mediate the TNF-alpha signaling pathway. Alternatively spliced transcript variants encoding different isoforms have been identified. [provided by RefSeq, Jul 2008]

Uniprot Description

HGK: a Ser/Thr kinase of the STE20 subfamily. May play a role in the response to environmental stress and cytokines such as TNF-alpha. May interact with the SH3 domain of the adapter proteins Nck. Binds, via its CNH regulatory domain, to the N- terminal region of SPG3A. Upregulated in primary tumors and tumor cell lines. Required for anchorage-independent growth and ras-dependent focus formation in multiple cell lines. 5 isoforms of the human protein are produced by alternative splicing.

Product Notes

The MAP4K4 map4k4 (Catalog #AAA6000261) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The NIK, CT (NF-kappa-beta-inducing Kinase, HsNIK, FTDCR1B, HS, Mitogen-activated Protein Kinase Kinase Kinase 14, MAP3K14, Serine/threonine Protein Kinase NIK) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's NIK can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Dilution: Western Blot: 1:1000-1:2000. Researchers should empirically determine the suitability of the MAP4K4 map4k4 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NIK, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
