Applications
ELISA, Immunohistochemistry, Western Blot
Synonyms
NESTIN; Polyclonal Antibody; RABBIT ANTI HUMAN NESTIN; anti-Nes antibody
Ordering
For Research Use Only!
Host
Rabbit
Reactivity
Bovine, Dog, Hamster, Macaque, Mouse, Rabbit, Rat
Clonality
Polyclonal
Isotype
IgG
Specificity
This item recognises human Nestin, an intermediate filament involved in cytoskeletal organisation.
Nestin is a marker of multipotent neuroectodermal cells and is downregulated as the cells cease division and differentiate to their neural or glial lineages.
Expression of Nestin is seen in almost all GBMs (Glioblastoma mulitformes) and many melanomas (both primary and metastatic) but not in any metastatic carcinomas.
This item has been reported to work in Immunofluorescence.
Form/Format
Purified
Purified IgG - liquid
Concentration
IgG concentration 0.5mg/ml (varies by lot)
Sequence Length
1864
Applicable Applications for anti-Nes antibody
ELISA (EIA), Immunohistology Paraffin*, Western Blot (WB)
Application Notes
ELISA: Minimum Dilution: 1/20000; Maximum Dilution: 1/80000;
Immunohistology - Paraffin: Maximum Dilution: 5ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Western Blotting: Minimum Dilution: 1/500; Maximum Dilution: 1/3000
Perservative Stabilisers
0.09% Sodium Azide (NaN3)
Immunogen
Recombinant human Nestin (aa1484-1500).
Histology Positive Control Tissue
Antiserum Preparation
Antisera to Human Nestin were raised by repeated immunisation of Rabbits with highly purified antigen. Purified IgG prepared from whole serum by affinity chromatography.
Target Species
Human
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

Uniprot Description

nestin: an intermediate filament protein. It is expressed predominantly in stem cells of the central nervous system in the neural tube. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments.

Protein type: Cytoskeletal

Cellular Component: cytoplasm; intermediate filament; intracellular

Molecular Function: protein binding; structural molecule activity; CCR5 chemokine receptor binding

Biological Process: nervous system development; multicellular organismal development; cell projection morphogenesis; negative regulation of neuron apoptosis

Product Notes

The Nes nes (Catalog #AAA224038) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI HUMAN NESTIN reacts with Bovine, Dog, Hamster, Macaque, Mouse, Rabbit, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's NESTIN can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistology Paraffin*, Western Blot (WB). ELISA: Minimum Dilution: 1/20000; Maximum Dilution: 1/80000; Immunohistology - Paraffin: Maximum Dilution: 5ug/ml; Application Note: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose. Western Blotting: Minimum Dilution: 1/500; Maximum Dilution: 1/3000. Researchers should empirically determine the suitability of the Nes nes for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "NESTIN, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
