Related Product Information for anti-KAT6A antibody
Histone acetyltransferases (HATs) have been implicated in a number of cellular functions including gene regulation, DNA synthesis, and repair. Histone acetyltransferases and deacetylases are respectively, the enzymes devoted to thr addition and removal of acetyl groups from lysine residues on the Histone N-terminal trails. The enzymes exert fundamental roles in developmental processes and their deregulation has been linked to the progression of diverse human disorders, including cancer.

NCBI Description

This gene encodes a member of the MOZ, YBFR2, SAS2, TIP60 family of histone acetyltransferases. The protein is composed of a nuclear localization domain, a double C2H2 zinc finger domain that binds to acetylated histone tails, a histone acetyl-transferase domain, a glutamate/aspartate-rich region, and a serine- and methionine-rich transactivation domain. It is part of a complex that acetylates lysine-9 residues in histone 3, and in addition, it acts as a co-activator for several transcription factors. Allelic variants of this gene are associated with autosomal dominant mental retardation-32. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Apr 2015]

Uniprot Description

Function: Histone acetyltransferase that acetylates lysine residues in histone H3 and histone H4 (in vitro). Component of the MOZ/MORF complex which has a histone H3 acetyltransferase activity. May act as a transcriptional coactivator for RUNX1 and RUNX2. Acetylates p53/TP53 at 'Lys-120' and 'Lys-382' and controls its transcriptional activity via association with PML. Ref.6 Ref.8 Ref.10 Ref.12 Ref.19 Ref.21

Catalytic activity: Acetyl-CoA + [histone] = CoA + acetyl-[histone]. Ref.6 Ref.7 Ref.21

Subunit structure: Component of the MOZ/MORF complex composed at least of ING5, KAT6A, KAT6B, MEAF6 and one of BRPF1, BRD1/BRPF2 and BRPF3. Interacts with RUNX1; phosphorylation of RUNX1 enhances the interaction. Interacts with RUNX2. Interacts with p53/TP53. Interacts with PML (isoform PML-4) and this interaction positively regulates its acetylation activity towards p53/TP53. Ref.6 Ref.8 Ref.12 Ref.14 Ref.19

Subcellular location: Nucleus. Nucleus › nucleolus. Nucleus › nucleoplasm. Nucleus › PML body. Note: Recruited into PML body after DNA damage. Ref.6 Ref.11 Ref.14

Domain: The N-terminus is involved in transcriptional activation while the C-terminus is involved in transcriptional repression. Ref.7

Post-translational modification: Autoacetylation at Lys-604 is required for proper function

By similarity. Autoacetylated.Phosphorylation at Thr-369 by PKB/AKT1 inhibits its interaction with PML and negatively regulates its acetylation activity towards p53/TP53.

Involvement in disease: Chromosomal aberrations involving KAT6A may be a cause of acute myeloid leukemias. Translocation t(8;16)(p11;p13) with CREBBP; translocation t(8;22)(p11;q13) with EP300. KAT6A-CREBBP may induce leukemia by inhibiting RUNX1-mediated transcription. Inversion inv8(p11;q13) generates the KAT6A-NCOA2 oncogene, which consists of the N-terminal part of KAT6A and the C-terminal part of NCOA2/TIF2. KAT6A-NCOA2 binds to CREBBP and disrupts its function in transcription activation.A chromosomal aberration involving KAT6A is a cause of therapy-related myelodysplastic syndrome. Translocation t(2;8)(p23;p11.2) with ASXL2 generates a KAT6A-ASXL2 fusion protein.

Sequence similarities: Belongs to the MYST (SAS/MOZ) family.Contains 1 C2HC-type zinc finger.Contains 1 H15 (linker histone H1/H5 globular) domain.Contains 2 PHD-type zinc fingers.

Product Notes

The KAT6A kat6a (Catalog #AAA646363) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The MYST3 (Histone Acetyltransferase KAT6A, MOZ, YBF2/SAS3, SAS2 and TIP60 Protein 3, MYST-3, Monocytic Leukemia Zinc Finger Protein, Runt-related Transcription Factor-binding Protein 2, Zinc Finger Protein 220, KAT6A, MOZ, RUNXBP2, ZNF220) reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's MYST3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Suitable for use in Western Blot. Dilution: Western Blot: 2-6ug/ml. Researchers should empirically determine the suitability of the KAT6A kat6a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MYST3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
