Application Notes
Suitable for use in Western Blot, Flow Cytometry and Direct ELISA.
Dilution: Western Blot: Can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse MUCDHL. The detection limit for rmMUCDHL is approximately 1 ng/lane under non-reducing and reducing conditions.
Flow Cytometry: Tested in flow cytometry using primary kidney cells. Dilute this antibody to 25ug/ml and add 10 L of the diluted solution to 1-2.5 x 10e5 cells in a total reaction volume not exceeding 200 L. The binding of unlabeled antibodies may be visualized by adding a secondary developing reagent such as anti-goat IgG conjugated to a fluorochrome.
Direct ELISA: Can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse MUCDHL. The detection limit for rmMUCDHL is approximately 0.5 ng/well.

Purified, NS0-derived rmMUCDHL (rmMUCDHL; aa 29 - 479; Accession # NP_082345)
Reconstitution
Reconstitute with sterile PBS. If 0.5ml of PBS is used, the antibody concentration will be 0.2mg/ml.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Mucin and cadherin-like protein (MUCDHL), also known as mu-protocadherin, is a 100kD type I transmembrane protein belonging to the protocadherin family of cell adhesion molecules. It is found apically on the brush border of renal proximal tubule epithelial cells. The extracellular domain of mature mouse MUCDHL contains four cadherin domains plus three repeat regions that constitute a mucin stalk (aa 541-614). At least one splice variant exists in mouse that shows a deletion of aa's 459-620. Over aa's 29-479, mouse MUCDHL shares 63% and 79% aa identity with human and rat MUCDHL, respectively.

NCBI Description

This gene is a novel mucin-like gene that is a member of the cadherin superfamily. While encoding nonpolymorphic tandem repeats rich in proline, serine and threonine similar to mucin proteins, the gene also contains sequence encoding calcium-binding motifs found in all cadherins. The role of the hybrid extracellular region and the specific function of this protein have not yet been determined. Alternatively spliced transcript variants encoding different isoforms have been described. [provided by RefSeq]
