Western Blot (WB)
(Western blot analysis of MGP expression in JURKAT whole cell lysates (lane 1). MGP at 12KD was detected using rabbit anti- MGP Antigen Affinity purified polyclonal antibody at 0.5 ug/mL. The blot was developed using chemiluminescence (ECL) method. )
Immunohistochemistry (IHC)
(MGP was detected in paraffin-embedded sections of human lung cancer tissues using rabbit anti- MGP Antigen Affinity purified polyclonal antibody at 1ug/mL. The immunohistochemical section was developed using SABC method. )
Background: Matrix Gla protein (MGP) is an 84-residue vitamin K-dependent protein initially isolated from bovine bone. In addition, MGP is a 10-kD protein produced and secreted by vascular smooth muscle cells and chondrocytes and significantly accumulated in bone, cartilage, and dentin. It is also expressed at high levels in heart, kidney, and lung and is upregulated by vitamin D in bone cells. MGP has a high affinity binding to calcium ions, similar to other Gla-containing proteins. The protein acts as an inhibitor of vascular mineralization and plays a role in bone organization. And this gene is mapped to 12p12.3.

NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is secreted and likely acts as an inhibitor of bone formation. The encoded protein is found in the organic matrix of bone and cartilage. Defects in this gene are a cause of Keutel syndrome (KS). Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jun 2010]
Uniprot Description
MGP: Associates with the organic matrix of bone and cartilage. Thought to act as an inhibitor of bone formation. Defects in MGP are the cause of Keutel syndrome (KS). KS is an autosomal recessive disorder characterized by abnormal cartilage calcification, peripheral pulmonary stenosis neural hearing loss and midfacial hypoplasia. Belongs to the osteocalcin/matrix Gla protein family.

Product Notes
The MGP mgp (Catalog #AAA178454) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-MGP Antibody reacts with Human No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's MGP can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin. Western Blot: 0.1-0.5mug/ml; Tested Species: Human Immunohistochemisry Paraffin: 0.5-1mug/ml; Tested Species: Human Tested Species:In-house tested species with positive results. By Heat: Boiling the paraffin sections in 10mM citrate buffer, pH6.0, for 20mins is required for the staining of formalin/paraffin sections. Other applications have not been tested. Researchers should empirically determine the suitability of the MGP mgp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MGP, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
