Description: Rabbit IgG polyclonal antibody for Methylated-DNA--protein-cysteine methyltransferase(MGMT) detection. Tested with WB, IHC-P, ICC in Human.

Background: O6-alkylguanine DNA alkyltransferase (also known as AGT, MGMT or AGAT) is a protein that in humans is encoded by the O6-methylguanine DNA methyltransferase (MGMT) gene. MGMT is mapped to 10q26.3. This gene is crucial for genome stability. It repairs the naturally occurring mutagenic DNA lesion O6-methylguanine back to guanine and prevents mismatch and errors during DNA replication and transcription. It has been found that methylation of the MGMT promoter in the tumor was associated with longer survival.

NCBI Description

Alkylating agents are potent carcinogens that can result in cell death, mutation and cancer. The protein encoded by this gene is a DNA repair protein that is involved in cellular defense against mutagenesis and toxicity from alkylating agents. The protein catalyzes transfer of methyl groups from O(6)-alkylguanine and other methylated moieties of the DNA to its own molecule, which repairs the toxic lesions. Methylation of the genes promoter has been associated with several cancer types, including colorectal cancer, lung cancer, lymphoma and glioblastoma. [provided by RefSeq, Sep 2015]

Uniprot Description

MGMT: Involved in the cellular defense against the biological effects of O6-methylguanine (O6-MeG) in DNA. Repairs alkylated guanine in DNA by stoichiometrically transferring the alkyl group at the O-6 position to a cysteine residue in the enzyme. This is a suicide reaction: the enzyme is irreversibly inactivated. Belongs to the MGMT family.

Product Notes

The MGMT mgmt (Catalog #AAA178332) is an Antibody and is intended for research purposes only. The product is available for immediate purchase. The Anti-MGMT Antibody reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's MGMT can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC) Paraffin, Immunocytochemistry (ICC). Western Blot Concentration: 0.1-0.5ug/ml Immunohistochemistry (IHC) Paraffin Concentration: 0.5-1ug/ml Immunocytochemistry Concentration: 0.5-1ug/ml. Researchers should empirically determine the suitability of the MGMT mgmt for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "MGMT, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
