Mitogen-activated protein kinase (MAPK) signaling cascades include MAPK or extracellular signal-regulated kinase (ERK), MAPK kinase (MKK or MEK), and MAPK kinase kinase (MAPKKK or MEKK). MAPKK kinase/MEKK phosphorylates and activates its downstream protein kinase, MAPK kinase/MEK, which in turn activates MAPK. The kinases of these signaling cascades are highly conserved, and homologs exist in yeast, Drosophila, and mammalian cells. MEKK1 can phosphorylate and activate MAPKK 1 and MAPKK 2 (MEK1/MEK2) which leads to phosphorylation of MAP kinases. It is also a highly efficient activator of the JNK cascade. The protein contains a putative 1 RING-type zinc finger and 1 SWIM-type zinc finger.

NCBI Description

The protein encoded by this gene is a serine/threonine kinase and is part of some signal transduction cascades, including the ERK and JNK kinase pathways as well as the NF-kappa-B pathway. The encoded protein is activated by autophosphorylation and requires magnesium as a cofactor in phosphorylating other proteins. This protein has E3 ligase activity conferred by a plant homeodomain (PHD) in its N-terminus and phospho-kinase activity conferred by a kinase domain in its C-terminus. MEKK1: a protein kinase of the STE11 family. Phosphorylates and activates MKK4 and -7, which in turn activate JNK1, -2 and -3. Contains 1 RING-type zinc finger. The RING finger domain of MEKK1 exhibits E3 ubiquitin ligase activity toward c-Jun. c-Jun is downregulated in response to osmotic stress via ubiquitination-mediated degradation by the RING finger domain of MEKK1. Overexpressed/overactivated in multiple tumor types. The Mek1/2 inhibitor U0126 blocks export of influenza viral particles and has been suggested as an antiviral treatment. Single mutations at the same point of each gene also associated with cardiofaciocutaneous syndrome, (also linked with Braf and Kras mutations), displaying morphological, cardiac and mental defects. Inhibitors: U0126, CI-1040/PD184352, PD-0325901 (Phase I cancer), ARRY-142886 (Phase 1, cancer).
