Application Notes
ELISA: Use at a concentration range of 0.5-2 ug/ml, detection limit 2-4 ng/ml using recombinant mouse CCL2
WB: Use at a concentration range of 0.1-0.2 ug/ml, detection limit 1.5-3 ng/lane using recombinant mouse CCL2
N: Use at a concentration range of 3-5 ug/ml to inhibit 50% (ND50)activity of recombinant mouse CCL2 at 100ng/ml
IHC: A suitable range of concentrations of this antibody for IHC is 2-10 ug/ml.

Polyclonal MCP-1 (CCL2) antibody was produced from sera of rabbits immunized with synthetic protein containing MCP-1 antigenic determinants.
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water. This solution can then be diluted into other buffers
Dilution
A quick spin of the vial followed by reconsititution in 0.01M PBS pH 7.4 or other diluents.
Preparation and Storage
This antibody can be stored in working aliquots at 2 degree C - 8 degree C for one month, or at -20 degree C for 12 months without detectable loss of activity.
Avoid repeated freeze/thaw cycles.

MCP-1 belongs to the family of chemotactic cytokines.MCP-1 is expressed by monocytes, vascular endothelial cells, smooth muscle cells, glomerular mesangial cell, osteoblastic cells. MCP-1 has been shown to exhibit biological activities other than Chemotaxis. It can induce the proliferation and activation of killer cells known as CHAK. Elevated levels of MCP-1 are observed in atherosclerotic plaques that are rich in macrophages. The factor activates the tumoricidal activity of monocytes and macrophages in vivo. It regulates the expression of cell surface antigens (CD11c, CD11b) and the expression of cytokines (IL1, IL6). MCP-1 is a potent activator of human basophils, inducing degranulation and the release of histamines. In basophils activated by IL3, IL5 or GM-CSF MCP-1 enhances the synthesis of leukotriene C4.

NCBI Description

This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4.
