Application Notes
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect human MBD-3. The detection limit for rhMBD-3 is approximately 5 ng/lane under non-reducing and reducing conditions., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human MBD-3. The detection limit for rhMBD-3 is approximately 0.5 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

MBD-3 (methyl-CpG-binding domain protein # 3) is a 38-40kD member of the MBD family of transcriptional repressors. Through its MBD, and as part of a large complex of regulatory proteins, it binds methylated DNA, promoting gene silencing during S phase. Although it can homodimerize, it is only active when heterodimerized with MBD-2. Human MBD-3 is 291 amino acids (aa) in length, and contains an N-terminal methyl binding domain (aa's 1-72) and a C-terminal coiled-coil region (aa's 216-245) that may be involved in protein-protein interaction. Human MBD-3 shares 95% aa identity with mouse, rat, and canine MBD-3, with differences essentially limited to the C-terminal 25 amino acids.

NCBI Description

DNA methylation is the major modification of eukaryotic genomes and plays an essential role in mammalian development. Human proteins MECP2, MBD1, MBD2, MBD3, and MBD4 comprise a family of nuclear proteins related by the presence in each of a methyl-CpG binding domain (MBD). However, unlike the other family members, MBD3 is not capable of binding to methylated DNA. The predicted MBD3 protein shares 71% and 94% identity with MBD2 (isoform 1) and mouse Mbd3. MBD3 is a subunit of the NuRD, a multisubunit complex containing nucleosome remodeling and histone deacetylase activities. MBD3 mediates the association of metastasis-associated protein 2 (MTA2) with the core histone deacetylase complex. [provided by RefSeq]

MBD3: methyl-CpG binding (MBD) proteins appear to bind differentially methylated domains (DMDs) of DNA, to recruit repression complexes, and to silence the locus. The methyl CpG binding domain of human MBD3 may not directly bind mCpG but does interact with the NuRD/Mi2 components HDAC1 and MTA2. Plays a role in histone deacetylation and nucleosome remodeling. Increased levels have been reported in lung cancer and glioblastomas and may be a therapeutic target.
