Dilution: Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4 degree C with gentle shaking, overnight
Immunogen
Synthetic phosphopeptide (KLH coupled) corresponding to residues surrounding Tyr507 of human Lyn.
Preparation and Storage
May be stored at 4 degree C for short-term only. For long-term storage, store at -20 degree C. Aliquots are stable for at least 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Lyn is a member of the src family of tyrosine kinases responsible for the regulation of cell signaling mechanisms. Lyn's function has been studied in cells of hematopoetic origin where it is associated with the high af nity IgE receptor in basophils and with the p120 Ras-GAP in thrombin-stimulated platelets. Lyn, one of the Src family members, is predominantly expressed in hematopoietic cells (1). Two tyrosyl residues have been reported to play a crucial role in the regulation of protein tyrosine kinases of the Src family: autophosphorylation of tyrosine 396 (equivalent to tyrosine 416 of c-Src) located in the catalytic domain correlates with enzyme activation, while Csk-mediated phosphorylation of the C-terminal tyrosine 507 (equivalent to tyrosine 527 of c-Src) inactives the kinases. Lyn was tyrosine phosphorylated and activated upon association with cell surface receptors such as the B cell Ag recepter (BCR) and CD40 (2-4). Knockout mice have shown that the net effect of Lyn deficiency is to render B cells hyperactive to BCR stimulation (5-7), suggesting that the most critical role for Lyn in vivo is in the downmodulation of B cell responses. Lyn is also involved in controlling the migration and development of specific B cell populations (8).
