Application Notes
Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse Latexin. The detection limit for rmLatexin is approximately 5 ng/lane and 0.5 ng/lane under non-reducing and reducing conditions, respectively., Immunoprecipitation: This antibody has been used to immunoprecipitate rmLatexin from conditioned media of transfected NS0 cells., Neutralization: This antibody has been used to neutralize the inhibitory activity of rmLatexin against rmCPA. The antibody was preincubated with the inhibitor at different molar ratios at room temperature for 30 minutes. The resulting mixtures were then incubated with the enzyme at room temperature for 60 minutes. The enzyme was assayed with a peptide substrate. 50% of the proteolytic activity was restored by the antibody at 4.4ug/ml (IC50) under conditions in which 0.1ug/ml of the inhibitor, 0.1ug/ml of the enzyme, and 100 M of the substrate were present. Considering the molecular masses of the inhibitor (26kD) and the antibody (150kD), IC50 was achieved at approximately 8:1 molar ratio of the antibody to the inhibitor., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse Latexin. The detection limit for rmLatexin is approximately 0.3 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
