WB: 1:1000 (when using colorimetric substrates such as BCIP/NBT)-1:5000 (for chemiluminescent substrates). Predicted molecular weight: 33 kDa. A number of different splice variants and partial sequences have been reported for Kallikrein 8, the longest encodes a protein of 305 amino acids with a predicted mass of 32.97 kDa. The 260 amino acid form has a predicted mass of 28 kDa. The 164, 134 and 119 amino acid versions have a predicted mass of 17.9, 14.7 and 12.7 kDa respectively but it is unclear if these proteins are produced. Dilution optimised using Chromogenic detection.

Preparation and Storage
This product is stable for several weeks at 4 degree C as an undiluted liquid. Dilute only prior to immediate use.

Description: The antibody is a rabbit polyclonal antibody made to the serine proteinase kallikrein-8 (hK8). The antibody is made to a synthetic peptide based on the catalytic domain of human Kallikrein-8. The antibody reacts with human Kallikrein-8, and does not react with the human kallikreins (hK1-15). The antibody has been peptide-affinity purified, concentrated to 1.0 mg/ml, with the addition of 0.05% sodium azide as preservative and 50% glycerol as cryoprotectant. Kallikrein-8 (hK8), also known as Neuropsin, Brain Serine Protease-1 (BSP1), tumor-associated differentially expressed gene-14 (TADG-14), PRSS19, HNP and Ovasin is a trypsin-like serine proteinase. The name neuopsin is intended to indicate a neuronal trypsin-like proteinase, but a literature alias for a form of opsin-5, also called neuopsin, confuses things a bit. Originally described from mouse brain, KLK8 was implicated in neural plasticity and learning. Kallikrein-8 is thought to be involved in neurite outgrowth and fasciculation during nervous system development. Kallikrein-8 is induced in neurite injury, and Kallikrein-8 has been shown to be elevated in the hippocampus on Alzheimer's patients' brains. Animal models of autoimmune encephalomyelitis (EAE) show elevated Kallikrein-8 production. Kallikrein-8 null mice have aberrant synapses in their hippocampus, compared to wild type mice, and are predisposed to seizures. Kallikrein-8 is thought to regulate the early phase of long-term potentiation (LTP), possibly by cleaving fibronectin and other substrates.Kallikrein-8 is also found in the uterus, where it is hormonally regulated. Cervical cancer cell lines express high levels of Kallikrein-8, and it may be useful as a tumor marker. A recommended starting concentration for Western blots is 1:1,000 when using colorimetric substrates such as BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for samples from more distantly related species.

NCBI Description

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in tandem in a gene cluster on chromosome 19. The encoded protein may be involved in proteolytic cascade in the skin and may serve as a biomarker for ovarian cancer. Alternate splicing of this gene results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2013]

KLK8: Serine protease which is capable of degrading a number of proteins such as casein, fibrinogen, kininogen, fibronectin and collagen type IV. Also cleaves L1CAM in response to increased neural activity. Induces neurite outgrowth and fasciculation of cultured hippocampal neurons. Plays a role in the formation and maturation of orphan and small synaptic boutons in the Schaffer- collateral pathway, regulates Schaffer-collateral long-term potentiation in the hippocampus and is required for memory acquisition and synaptic plasticity. Involved in skin desquamation and keratinocyte proliferation. Plays a role in the secondary phase of pathogenesis following spinal cord injury. Belongs to the peptidase S1 family. Kallikrein subfamily. 4 isoforms of the human protein are produced by alternative splicing.
