Application Notes
ELISA: This purified human IL-8 antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with a biotinylated human IL-8 antibody as the detection reagent and recombinant human IL-8 as the standard.
Western Blot: This purified human IL-8 antibody may used in Western Blotting applications under either reducing or non-reducing conditions with recombinant human IL-8 as the positive control.
ELISA: Minimum Dilution: 0.5; Maximum Dilution: 2.0ug/ml
Functional Assays: Minimum Dilution: 2.0; Maximum Dilution: 3.0ug/ml
Immunohistology - Paraffin: Minimum Dilution: 0.25; Maximum Dilution: 1.0ug/ml; Application Note: This product requires enzyme mediated antigen retrieval prior to staining of parrafin sections. Proteinase K is recomended for this purpose.
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution. For long term storage the addition of 0.09% sodium azide is recommended. N.B. For functional studies do not add sodium azide.

Preparation and Storage
Prior to reconstitution store at 4 degree C. Following reconstitution store at -20 degree C. This product should be stored undiluted. Storage in frost-free freezers is not recommended. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti Human Interleukin-8 antibody recognizes human IL-8 (Interleukin-8), a 72 amino acid pro-inflammatory CXC chemokine designated CXCL8, secreted by endothelial cells and monocytes, which acts as a major chemoattractant for neutrophils and also T cells and basophils. IL-8, first isolated from osteosarcoma cells, contains the ELR-motif (N-Terminal Glu-Leu-Arg amino acid sequence) and signals through the CXCR1 and CXCR2 receptors. Cancer studies have demonstrated a role for IL-8 in the angiogenesis and growth of tumours. Previous nomenclature for IL-8 includes neutrophil activating protein 1(NAP-1), granulocyte chemotactic protein 1 (GCP-1), monocyte-derived neutrophil-activating peptide (MONAP) and protein 3-10C.

NCBI Description

The protein encoded by this gene is a member of the CXC chemokine family. This chemokine is one of the major mediators of the inflammatory response. This chemokine is secreted by several cell types. It functions as a chemoattractant, and is also a potent angiogenic factor. This gene is believed to play a role in the pathogenesis of bronchiolitis, a common respiratory tract disease caused by viral infection. This gene and other ten members of the CXC chemokine gene family form a chemokine gene cluster in a region mapped to chromosome 4q. [provided by RefSeq, Jul 2008]

IL8: IL-8 is a chemotactic factor that attracts neutrophils, basophils, and T-cells, but not monocytes. It is also involved in neutrophil activation. It is released from several cell types in response to an inflammatory stimulus. IL-8(6-77) has a 5-10-fold higher activity on neutrophil activation, IL-8(5-77) has increased activity on neutrophil activation and IL-8(7-77) has a higher affinity to receptors CXCR1 and CXCR2 as compared to IL-8(1-77), respectively. Belongs to the intercrine alpha (chemokine CxC) family. 2 isoforms of the human protein are produced by alternative splicing.
