Western Blot (WB)
(Detection of Human MERIT40 by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line. Gel was loaded with 20ug of cytoplasmic (Cyto) and 10 μg of nuclear extracts (Nuc). PVDF Membrane was probed with 1ug/ml of Human MERIT40 Antigen Affinitypurified Polyclonal Antibody followed by HRPconjugated Sheep IgG Secondary Antibody. A specific band was detected for MERIT40 at approximately 40kD (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

(MERIT40 in Human Breast Cancer Tissue. MERIT40 was detected in immersion fixed paraffinembedded sections of human breast cancer tissue using Human MERIT40 Antigen Affinitypurified Polyclonal Antibody at 10ug/ml overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heatinduced epitope retrieval using Antigen Retrieval ReagentBasic. Tissue was stained using the Sheep HRPDAB Cell & Tissue Staining Kit a n d counterstained with hematoxylin (blue). Specific staining was localized to glandular epithelial cells. Function: Component of the BRCA1-A complex, a complex that specifically recognizes 'Lys-63'-linked ubiquitinated histones H2A and H2AX at DNA lesions sites, leading to target the BRCA1-BARD1 heterodimer to sites of DNA damage at double-strand breaks (DSBs). The BRCA1-A complex also possesses deubiquitinase activity that specifically removes 'Lys-63'-linked ubiquitin on histones H2A and H2AX. In the BRCA1-A complex, it is required for the complex integrity and its localization at DSBs. Probably also plays a role as a component of the BRISC complex, a multiprotein complex that specifically cleaves 'Lys-63'-linked ubiquitin. In these 2 complexes, it is probably required to maintain the stability of BRE/BRCC45 and help the 'Lys-63'-linked deubiquitinase activity mediated by BRCC3/BRCC36 component. Ref.12 Ref.13 Ref.14
