Western Blot (WB) (Detection of Human MERIT40 by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line. Gel was loaded with 20ug of cytoplasmic (Cyto) and 10 μg of nuclear extracts (Nuc). PVDF Membrane was probed with 1ug/ml of Human MERIT40 Antigen Affinity­purified Polyclonal Antibody followed by HRP­conjugated Sheep IgG Secondary Antibody. A specific band was detected for MERIT40 at approximately 40kD (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

Recombinant corresponding to aa182-329 from human MERIT40 expressed in E. coli (Q9NWV8).
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Immunohistochemistry (IHC)

MERIT40 was detected in immersion fixed paraffin­embedded sections of human breast cancer tissue using Human MERIT40 Antigen Affinity­purified Polyclonal Antibody at 10ug/ml overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat­induced epitope retrieval using Antigen Retrieval Reagent­Basic. Tissue was stained using the Sheep HRP­DAB Cell & Tissue Staining Kit a n d counterstained with hematoxylin (blue). Specific staining was localized to glandular epithelial cells. MERIT40 is a 40kD member of the MERIT40 family of proteins. It is ubiquitously expressed, and participates in the DNA damage response. At sites of double strand breaks, histones are phosphorylated, ubiquitinated, and deubiquitinated, with repairs performed by a sequentially formed complex that contains BRCA1, Rap80 and BRE. MERIT40 apparently serves as a scaffold protein that maintains the integrity of the complex. Human MERIT40 is 329aa in length. It contains one VWF-A domain aa95-298 and six utilized Ser/Thr phosphorylation sites. There are three potential splice variants. One shows a 13aa substitution for aa234-329, a second possesses a 105aa substitution for aa263-329, while a third shows a 37aa substitution for aa1-115, coupled to a deletion of aa263-329. Over aa182-329, human MERIT40 shares 95aa identity with mouse MERIT40.

Uniprot Description

Function: Component of the BRCA1-A complex, a complex that specifically recognizes 'Lys-63'-linked ubiquitinated histones H2A and H2AX at DNA lesions sites, leading to target the BRCA1-BARD1 heterodimer to sites of DNA damage at double-strand breaks (DSBs). The BRCA1-A complex also possesses deubiquitinase activity that specifically removes 'Lys-63'-linked ubiquitin on histones H2A and H2AX. In the BRCA1-A complex, it is required for the complex integrity and its localization at DSBs. Probably also plays a role as a component of the BRISC complex, a multiprotein complex that specifically cleaves 'Lys-63'-linked ubiquitin. In these 2 complexes, it is probably required to maintain the stability of BRE/BRCC45 and help the 'Lys-63'-linked deubiquitinase activity mediated by BRCC3/BRCC36 component. Ref.12 Ref.13 Ref.14

Subunit structure: Component of the BRCA1-A complex, at least composed of the BRCA1, BARD1, UIMC1/RAP80, FAM175A/Abraxas, BRCC3/BRCC36, BRE/BRCC45 and BABAM1/NBA1. In the BRCA1-A complex, interacts directly with FAM175A/Abraxas and BRE/BRCC45. Component of the BRISC complex, at least composed of the FAM175B/ABRO1, BRCC3/BRCC36, BRE/BRCC45 and BABAM1/NBA1. Domain: The VWFA-like region is similar to the VWFA domain. Its presence reveals similarities between the structure of the 19S proteasome and the BRCA1-A complexes. Ref.13
