Dilution: Peptide Inhibition: 10uM of a histone H4 peptide containing acetyl-lysine 12 abolished detection of histone H4 by anti-acetyl-Histone H4 (Lys12) in immunoblot analysis of HeLa acid extracts. Little to no signal reduction was observed with 10uM of histone H4 peptides containing acetyl-lysine 5, acetyl-lysine 8 or acetyl-lysine 16.
Immunoprecipitation: 2-6ul immunoprecipitated the Ime2 promoter from wild type yeast chromatin and not from mutant yeast chromatin containing a histone H4 K12R mutation.
Western Blot: 1:3,000 to 1:10,000 detects acetyl-Histone H4 (Lys12) in acid extracted proteins from HeLa cells. The antibody did not detect unacetylated recombinant Histone H4.

Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails; instead, they contain a palindromic termination element. [provided by RefSeq]
