NCBI Description

The product encoded by this gene is involved in the maintenance of iron homeostasis, and it is necessary for the regulation of iron storage in macrophages, and for intestinal iron absorption. The preproprotein is post-translationally cleaved into mature peptides of 20, 22 and 25 amino acids, and these active peptides are rich in cysteines, which form intramolecular bonds that stabilize their beta-sheet structures. These peptides exhibit antimicrobial activity against bacteria and fungi. Mutations in this gene cause hemochromatosis type 2B, also known as juvenile hemochromatosis, a disease caused by severe iron overload that results in cardiomyopathy, cirrhosis, and endocrine failure. [provided by RefSeq, Oct 2014]

Uniprot Description

HAMP: Seems to act as a signaling molecule involved in the maintenance of iron homeostasis. Seems to be required in conjunction with HFE to regulate both intestinal iron absorption and iron storage in macrophages. Defects in HAMP are the cause of hemochromatosis type 2B (HFE2B); also known as juvenile hemochromatosis (JH). HFE2B is a disorder of iron metabolism with excess deposition of iron in the tissues, bronze skin pigmentation, hepatic cirrhosis, arthropathy and diabetes. The most common symptoms of hemochromatosis type 2 at presentation are hypogonadism and cardiomyopathy. Belongs to the hepcidin family.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 19q13.1

Cellular Component: apical cortex; cell; extracellular region; extracellular space

Molecular Function: hormone activity; receptor binding

Biological Process: cellular iron ion homeostasis; defense response to bacterium; defense response to fungus; immune response; killing of cells of another organism; negative regulation of ion transmembrane transporter activity; negative regulation of transcription from RNA polymerase II promoter; positive regulation of receptor internalization; response to iron ion

Disease: Hemochromatosis, Type 2b

Product Notes

The HAMP hamp (Catalog #AAA421783) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The Goat anti-HAMP Antibody reacts with Expected from sequence similarity: Human and may cross-react with other species as described in the data sheet. AAA Biotech's HAMP can be used in a range of immunoassay formats including, but not limited to, Peptide ELISA (EIA). Peptide ELISA: Antibody detection limit dilution 1: 16000. Western Blot: Preliminary experiments gave an approx 28kDa band in Human Liver lysates after 0.1ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 9.41kDa according to NP_066998.1. The 28kDa band was successfully blocked by incubation with the immunizing peptide. Researchers should empirically determine the suitability of the HAMP hamp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: TSGSVFPQQT GQLAE. It is sometimes possible for the material contained within the vial of "HAMP, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
