Immunohistochemistry (IHC)

(Immunohistochemical analysis of GRP75 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

Immunofluorescence (IF)

(Immunofluorescent analysis of GRP75 staining in NIH3T3 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.)

NCBI Description

This gene encodes a member of the heat shock protein 70 gene family. The encoded protein is primarily localized to the mitochondria but is also found in the endoplasmic reticulum, plasma membrane and cytoplasmic vesicles. This protein is a heat-shock cognate protein. This protein plays a role in cell proliferation, stress response and maintenance of the mitochondria. A pseudogene of this gene is found on chromosome 2.[provided by RefSeq, May 2010]

Uniprot Description

HSPA9B: Implicated in the control of cell proliferation and cellular aging. May also act as a chaperone. Interacts with FXN. Interacts with HSCB. Component of the MINOS/MitOS complex, that includes IMMT, HSPA9 and CHCHD3 and associates with mitochondrial outer membrane proteins SAMM50, MTX1 and MTX2. Belongs to the heat shock protein 70 family.

Protein type: Heat shock protein; Chaperone; Nucleolus

Chromosomal Location of Human Ortholog: 5q31.1

Cellular Component: focal adhesion; mitochondrion; cytoplasm; nucleolus

Molecular Function: protein binding; ubiquitin protein ligase binding; unfolded protein binding; ATP binding

Biological Process: cellular protein metabolic process; protein folding; protein export from nucleus; protein targeting to mitochondrion; negative regulation of apoptosis

Product Notes

The HSPA9 hspa9 (Catalog #AAA821824) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-GRP75 Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's GRP75 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). WB (1/500 - 1/1000), IHC (1/100 - 1/200), IF/ICC (1/100 - 1/500). Researchers should empirically determine the suitability of the HSPA9 hspa9 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GRP75, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
