Application Notes
ICC: Cells were fixed using 4% formaldehyde in PBS, pH 7.4 for 20 minutes at RT. The cells are permeabilized with 0.2% Triton X-100 and 2% serum (corresponding to the animal in which the secondary antibody was raised) in PBS for 20 minutes at RT. Cell cultures are then incubated with primary antisera in PBS containing 2% serum (as above), at 4 degree C, overnight. Has also been used successfully with the avidin/biotin/peroxidase detection system. Cultures were washed four times (10 minutes each) between steps, and reaction product is developed with a 0.05% diaminobenzidine/ 0.02% hydrogen peroxide solution on ice for 8-10 minutes.
Applications are based on unconjugated antibody.

Preparation and Storage
Store product at 4 degree C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20 degree C. Aliquots are stable at -20 degree C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Uniprot Description

Energy-transfer acceptor. Its role is to transduce the blue chemiluminescence of the protein aequorin into green fluorescent light by energy transfer. Fluoresces in vivo upon receiving energy from the Ca2+-activated photoprotein aequorin.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Green Fluorescent Protein, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
