Predicted Reactivity: Pig(83%), Horse(100%), Dog(100%)
IF: 1:100-1:500
ICC: 1:100-1:500
ELISA(peptide): 1:20,000-1:40,000
Western Blot (WB)
(Western blot analysis of extracts from K562, using FLIP delta + gamma Antibody. The lane on the left was treated with blocking peptide.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612492 staining A549 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612492) and mouse anti-beta tubulin Ab(T0023) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Post Translational Modifications: Proteolytically processed by CASP8 generating subunit p43 and p12.
Tissue Specificity: Widely expressed. Higher expression in skeletal muscle, pancreas, heart, kidney, placenta, and peripheral blood leukocytes. Also detected in diverse cell lines. Isoform 8 is predominantly expressed in testis and skeletal muscle.
Subunit Structure: TNFRSF6 stimulation triggers recruitment to the death-inducing signaling complex (DISC) formed by TNFRSF6, FADD and CASP8 (PubMed:9880531). A proteolytic fragment (p43) stays associated with the DISC (PubMed:9880531). Also interacts with FADD, CASP8, CASP3, TRAF1, TRAF2 and Bcl-X(L) (in vitro) (PubMed:9326610, PubMed:9208847). Interacts with RIPK1 (By similarity) (PubMed:9326610, PubMed:9208847, PubMed:9880531).(Microbial infection) Interacts with HBV protein X.
Similarity: The caspase domain lacks the active site residues involved in catalysis.Belongs to the peptidase C14A family.

Predicted Molecular Weight: (Calculated)55kDa.
NCBI Description
The protein encoded by this gene is a regulator of apoptosis and is structurally similar to caspase-8. However, the encoded protein lacks caspase activity and appears to be itself cleaved into two peptides by caspase-8. Several transcript variants encoding different isoforms have been found for this gene, and partial evidence for several more variants exists. [provided by RefSeq, Feb 2011]

Product Notes
The CFLAR cflar (Catalog #AAA9612492) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The FLIP Antibody reacts with Human Predicted Reactivity: Pig(83%), Horse(100%), Dog(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's FLIP delta + gamma can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the CFLAR cflar for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "FLIP delta + gamma, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
