Immunofluorescence (IF)

Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human FBXO28 polyclonal antibody (dilution ratio 1:1000) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus.)

The F-box protein is the substrate recognition subunit of SCF (SKP1/CUL1/F-box) E3 ubiquitin ligase complex, a multicomponent RING-type E3 ligase involved in the regulation of numerous cellular processes by targeting critical regulatory proteins for ubiquitination. F-box protein 28 (FBXO28), a substrate-recruiting component of the Skp1-Cul1-F-box (SCF) ligase complex, is a regulator of pancreatic beta-cell survival. FBXO28 is an important nuclear F-box protein. FBXO28 is involved in cell cycle regulation and required for appropriate mitotic progression. Loss of FBXO28 results in metaphase to anaphase progression delay, which then leads to several mitotic defects like lagging chromosomes, multipolar spindles and multi-nucleation.
