Estrogen related receptor alpha (ERRalpha) is a nuclear receptor in the superfamily of ligand-regulated transcription factors and is a member of the NR3B orphan nuclear receptor subgroup (consisting of alpha, beta and gamma). ERRalpha plays a role in modulating the estrogen signaling pathway. In addition, the expression of ERRalpha has been shown to increase during fasting and cold exposure. ERRalpha may be important for regulating mitochondrial biogenesis and oxidative metabolism by acting directly on genes necessary for mitochondrial function. Mice lacking ERRalpha are unable to maintain their body temperature in the cold. ERRalpha may also be involved in the maintenance and formation of cartilage. This information could be useful in finding therapeutic agents for a variety of diseases affecting the joints.

NCBI Description

The protein encoded by this gene is a nuclear receptor that is most closely related to the estrogen receptor. This protein acts as a site-specific transcription factor and interacts with members of the PGC-1 family of transcription cofactors to regulate the expression of most genes involved in cellular energy production as well as in the process of mitochondrial biogenesis. A processed pseudogene of ESRRA is located on chromosome 13q12.1. [provided by RefSeq, Jun 2019]

ERR1: Binds to an ERR-alpha response element (ERRE) containing a single consensus half-site, 5'-TNAAGGTCA-3'. Can bind to the medium-chain acyl coenzyme A dehydrogenase (MCAD) response element NRRE-1 and may act as an important regulator of MCAD promoter. Binds to the C1 region of the lactoferrin gene promoter. Requires dimerization and the coactivator, PGC-1A, for full activity. The ERRalpha/PGC1alpha complex is a regulator of energy metabolism. Belongs to the nuclear hormone receptor family. NR3 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
