Rabbit ERBB2 Polyclonal Antibody | anti-ERBB2 antibody
ERBB2 Antibody (N-term)
Western Blot (WB)
(Western blot analysis of ErbB2 (arrow) in T47D cell lysates, either noninduced (Lane 1) or induced with HRG (Lane 2).)
Flow Cytometry (FC/FACS)
(Flow cytometric analysis of MCF-7 cells using HER2/ErbB2 Antibody (N-term) (bottom histogram) compared to a negative control cell (top histogram). FITC-conjugated goat-anti-rabbit secondary antibodies were used for the analysis.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by AEC staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.)
Immunohistochemistry (IHC)
(Formalin-fixed and paraffin-embedded human breast carcinoma tissue reacted with HER2/ErbB2 antibody (N-term) , which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.)

NCBI and Uniprot Product Information
NCBI Description
This gene encodes a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. This protein has no ligand binding domain of its own and therefore cannot bind growth factors. However, it does bind tightly to other ligand-bound EGF receptor family members to form a heterodimer, stabilizing ligand binding and enhancing kinase-mediated activation of downstream signalling pathways, such as those involving mitogen-activated protein kinase and phosphatidylinositol-3 kinase. Allelic variations at amino acid positions 654 and 655 of isoform a (positions 624 and 625 of isoform b) have been reported, with the most common allele, Ile654/Ile655, shown here. Amplification and/or overexpression of this gene has been reported in numerous cancers, including breast and ovarian tumors. Alternative splicing results in several additional transcript variants, some encoding different isoforms and others that have not been fully characterized. [provided by RefSeq, Jul 2008]
Uniprot Description
HER2: a proto-oncogenic receptor tyrosine kinase of the EGFR family. Essential component of a neuregulin-receptor complex, although neuregulins do not interact with it alone. Not activated by EGF, TGF- alpha and amphiregulin. Amplified in breast cancer. Overexpression induces constitutive activity, and the gene is amplified or overexpressed in up to 30% of breast cancers, correlating with poor survival. The antibody Herceptin is approved for treatment of metastatic breast cancer with HER2 amplification/overexpression. Somatic mutations seen in 4% of lung cancers and also in breast, gastric, ovarian cancer and glioblastoma. One SNP shows predisposition to breast and gastric cancer. Inhibitors: Herceptin, lapatinib, PKI-166, EKB-569, CI-1033.

Product Notes
The ERBB2 erbb2 (Catalog #AAA9207444) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 21-52. The ERBB2 Antibody (N-term) reacts with Human (Predicted Reactivity: Mouse, Rat) and may cross-react with other species as described in the data sheet. AAA Biotech's ERBB2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA (EIA), Flow Cytometry (FC/FACS), Immunohistochemistry (IHC). WB~~1:1000. Researchers should empirically determine the suitability of the ERBB2 erbb2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ERBB2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
