eIF4A1 is a 406aa ATP-dependent RNA helicase belonging to the DEAD box helicase family and eukaryotic initiation factor-4A subfamily with a helicase ATP-binding domain and a helicase C-terminal domain. eIF4F eukaryotic translation initiation complex is formed by the association of eIF4A1 with eIF4E and eIF4E kinase (MNK1), eIF4B, eIF4G, PABPC1. It is closely related to eIF4A2. eIF4A1 is a eukaryotic translation initiation factor involved in protein translation:synthesis where it helps in unwinding the secondary structure of RNA in the 5'-UTR of mRNAs, cap recognition, and is necessary for efficient binding mRNA to the 40S small ribosomal subunit, as well as subsequent scanning for the initiator codon. Ubiquitous expression of the enzyme eIF4A1 is seen in most tissues.

Uniprot Description

EIF4A1: ATP-dependent RNA helicase which is a subunit of the eIF4F complex involved in cap recognition and is required for mRNA binding to ribosome. In the current model of translation initiation, eIF4A unwinds RNA secondary structures in the 5'-UTR of mRNAs which is necessary to allow efficient binding of the small ribosomal subunit, and subsequent scanning for the initiator codon. eIF4F is a multi-subunit complex, the composition of which varies with external and internal environmental conditions. It is composed of at least EIF4A, EIF4E and EIF4G1/EIF4G3. Interacts with PAIP1, EIF4E and UPF2. Found in a complex with XPO7, EIF4A1, ARHGAP1, VPS26A, VPS29, VPS35 and SFN. May interact with NOM1. Interacts with PDCD4; this interferes with the interaction between EIF4A and EIF4G. Interacts with RBM4. Interacts with human cytomegalovirus/HHV-5 protein UL69. Helicase activity and function in translation are inhibited by interaction with PDCD4. Belongs to the DEAD box helicase family. eIF4A subfamily.
