Related Product Information for anti-DNMT1 antibody
Methylation of DNA at cytosine residues plays an important role in regulation of gene expression, genomic imprinting and is essential for mammalian development. Hypermethylation of CpG islands in tumor suppressor genes may be linked with development of cancer. Three families of mammalian DNA methyltransferase genes have been identified which include Dnmt1, Dnmt2 and Dnmt3. Dnmt1 is constitutively expressed in proliferating cells and inactivation of Dnmt1 causes global demethylation of genomic DNA and embryonic lethality. Dnmt1 co-purifies with the retinoblastoma (Rb) tumor suppressor gene product, E2F1, and HDAC1. Dnmt1 also cooperates with Rb to repress transcription from promoters containing E2F binding sites suggesting a link between DNA methylation, histone deacetylase and sequence-specific DNA binding activity, as well as a growth-regulatory pathway that is disrupted in nearly all cancer cells. Phosphorylation of Ser714 is induced by EGF, but the role of this modification is unknown.

NCBI Description

DNA (cytosine-5-)-methyltransferase 1 has a role in the establishment and regulation of tissue-specific patterns of methylated cytosine residues. Aberrant methylation patterns are associated with certain human tumors and developmental abnormalities. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]

Uniprot Description

DNMT1: an ubiquitous DNA methyltransferase that methylates CpG residues. Preferentially methylates hemimethylated DNA. It is responsible for maintaining methylation patterns established in development, and may play an active role in DNA damage repair. Mediates transcriptional repression by direct binding to HDAC2. Its abundance is reduced to non detectable levels at the G0 phase of the cell cycle and is dramatically induced upon entrance into the S-phase of the cell cycle. Interacts with HDAC1 and with PCNA. Forms a complex with DMAP1 and HDAC2, with direct interaction. Forms also a stable complex with E2F1, BB1 and HDAC1. Binds MBD2 and MBD3. Three isoforms of the human protein produced by alternative splicing have been described.

Product Notes

The DNMT1 dnmt1 (Catalog #AAA629513) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The DNA Methyltransferase 1, phosphorylated (Ser714) (Dnmt 1, MCMT, M.Hsa1, DNA cytosine 5 methyltransferase 1, DNA MTase Mmu1, DNA methyltransferase M.HsaI) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's DNA Methyltransferase 1 can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Gel Shift Assay (GS/EMSA). Suitable for use in ELISA, Western Blot, and Dot Blot. Dilution: ELISA: 1:1000 Western Blot: 1:250 Dot Blot: 1:500. Researchers should empirically determine the suitability of the DNMT1 dnmt1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DNA Methyltransferase 1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
