Western Blot (WB) (Figure 1. Western blot analysis of Cystatin C using anti-Cystatin C antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: mouse brain tissue lysates,Lane 2: mouse spleen tissue lysates,Lane 3: mouse heart tissue lysates,Lane 4: mouse lung tissue lysates,Lane 5: mouse kidney tissue lysates,Lane 6: mouse testis tissue lysates,Lane 7: mouse HEPA1-6 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Cystatin C antigen affinity purified polyclonal antibody at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Cystatin C at approximately 16KD. The expected band size for Cystatin C is at 16KD.)

Related Product Information for anti-Cst3 antibody
Description: Rabbit IgG polyclonal antibody for Cystatin C detection. Tested with WB, ELISA (Cap) in Mouse; Rat.
Background: Cystatin C or cystatin 3, a protein encoded by the CST3 gene, is mainly used as a biomarker of kidney function. Recently, it has been studied for its role in predicting new-onset or deteriorating cardiovascular disease. It also seems to play a role in brain disorders involving amyloid, such as Alzheimer's disease. In humans, all cells with a nucleus (cell core containing the DNA) produce cystatin C as a chain of 120 amino acids. It is found in virtually all tissues and body fluids. It is a potent inhibitor of lysosomal proteinases (enzymes from a special subunit of the cell that break down proteins) and probably one of the most important extracellular inhibitors of cysteine proteases (it prevents the breakdown of proteins outside the cell by a specific type of protein degrading enzymes). Cystatin C belongs to the type 2 cystatin gene family.

NCBI Description

The protein encoded by this gene is a cysteine protease inhibitor involved in neurodegenerative and cardiovascular processes. The encoded protein inhibits aggregation of beta-amyloid protein, a hallmark of Alzheimer's disease, so it may be useful as a therapeutic. This protein also may be a biomarker for atherosclerosis. [provided by RefSeq, Aug 2015]

Uniprot Description

As an inhibitor of cysteine proteinases, this protein is thought to serve an important physiological role as a local regulator of this enzyme activity.

Product Notes

The Cst3 cst3 (Catalog #AAA1751291) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Cystatin C/Cst3 Antibody reacts with Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Cystatin C/Cst3 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), ELISA Capture (EIA). WB: 0.1-0.5 mug/ml ELISA (Cap):1-5 mug/ml Tested Species: In-house tested species with positive results. Researchers should empirically determine the suitability of the Cst3 cst3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Cystatin C/Cst3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
