Immunohistochemistry (IHC)

(Immunohistochemical analysis of CTBP2 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

Immunofluorescence (IF)

(Immunofluorescent analysis of CTBP2 staining in A549 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.)

NCBI Description

This gene produces alternative transcripts encoding two distinct proteins. One protein is a transcriptional repressor, while the other isoform is a major component of specialized synapses known as synaptic ribbons. Both proteins contain a NAD+ binding domain similar to NAD+-dependent 2-hydroxyacid dehydrogenases. A portion of the 3' untranslated region was used to map this gene to chromosome 21q21.3; however, it was noted that similar loci elsewhere in the genome are likely. Blast analysis shows that this gene is present on chromosome 10. Several transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Feb 2014]

Uniprot Description

CTBP2: Corepressor targeting diverse transcription regulators. Functions in brown adipose tissue (BAT) differentiation. Belongs to the D-isomer specific 2-hydroxyacid dehydrogenase family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Nuclear receptor co-regulator; Transcription, coactivator/corepressor

Chromosomal Location of Human Ortholog: 10q26.13

Cellular Component: transcriptional repressor complex; synapse; cell junction; nucleus

Molecular Function: protein binding; retinoic acid receptor binding; transcription coactivator activity; chromatin binding; NAD binding; transcription corepressor activity; oxidoreductase activity, acting on the CH-OH group of donors, NAD or NADP as acceptor

Biological Process: negative regulation of cell proliferation; transcription, DNA-dependent; white fat cell differentiation; positive regulation of transcription from RNA polymerase II promoter; positive regulation of retinoic acid receptor signaling pathway; viral genome replication; negative regulation of transcription, DNA-dependent

Product Notes

The CTBP2 ctbp2 (Catalog #AAA8225776) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-CTBP2 Antibody reacts with Human, Mouse, Rat, Bovine and may cross-react with other species as described in the data sheet. AAA Biotech's CTBP2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB),Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). WB (1/500 - 1/1000), IHC (1/100 - 1/200), IF (1/100 - 1/500), ICC (1/100 - 1/500). Researchers should empirically determine the suitability of the CTBP2 ctbp2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CTBP2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
